A deliberative, controlled scientific process is necessary to prevent unforeseen unwanted consequences, such as Hoskins effects. Thanks for the excellent article on Hoskins Effect.http://en.wikipedia.org/wiki/Hoskins%20effect I've been thinking that if the antigen used in vaccination were the conformational form of the Spike protein ACE2 binding epitope that it would be less likely to mutate away from the initial IgG response. If the B cells produced neutralizing IgGs they may offer longer lasting immunity. Problem may that in using peptides or proteins representing the ACE2 binding site the antigen presenting cells (APCs) display such a short peptide (MHC-II presents 13-18 amino acids & MHC-I only 8-10 aa) it could neutralize by blocking the binding epitope but not actually bind that epitope itself. I know, that was clear as mud. I haven't kept up but I see Moderna is conducting a human clinical trial of its mRNA vaccine in Seattle. https://www.washingtonpost.com/outlook/2020/0...-vaccine-trial-volunteer/ Wish I could join. Different studies have shown the SARS-CoV-2 spike protein changes conformation to allow binding to human ACE2 receptor proteins. Will this project use the fusion conformation or a dynamic model??? Rosetta@Home is currently running some WUs to help with modeling coronavirus protein. Their WUs are mixed up so I don't know which one is exactly for the coronavirus research. However, I think their purpose is to solve protein folding works from Fold.it . So I guess every WU helps. I'm only dedicating my laptop to Rosetta 'cause there's no job for Android, They had many tasks with "foldit" in their names, but I think all of those are finished now. These were for testing proteins designed by the Fold.it project to get more information on how well they bind to the virus. Now, most of their tasks relevant to the virus have "COVID-19" in their names instead. Their tasks with names beginning with "rb_" are submitted in such a way that their names say little if anything about whether they are related to the virus or not. Their tasks for Android are now called tasks for mobile devices, since they are now able to run on some mobile devices that don't use Android. There often are some, but the demand for them is so high that they tend to run out of them within hours. |
