Experiment #1 which was our initial screen of the NCI diversity set against 270 protease variants has been analyzed and the first cut at promising leads was identified. We selected the top 40 of these compounds and ordered them from the National Cancer Institute. They arrived yesterday and they have been delivered to our collaborators here at Scripps for experimental assay. They will first be assayed against wild-type HIV protease. Subsequently they will be tested against a number of HIV protease mutants that are available. We anxiously await the first results from these assays.

Many thanks for the update! Just wondering, are these some kind of Cancer compound? I wouldn't think so, but because they were ordered at the National Cancer Institute?

I think the NCI functions as central ripository of all compounds known and stocked, which medical research labs can request. Maybe there is also a quality control function in there to ensure, that all labs work with the same compound when they say to work with compound X.

... are these some kind of Cancer compound? ... Are these compounds tested "in real life"? I mean, not virtual? Not necessarily cancer compounds. From the web site of the NCI's DTP (that supplies the compounds): "As the drug discovery and development arm of the National Cancer Institute, the Developmental Therapeutics Program (DTP) plans, conducts, and facilitates development of therapeutic agents for cancer and AIDS." Yes, when we say "experimental assay" that means an experiment in the lab in which the real compounds are assayed for their ability to inhibit the enzyme activity of the purified (i.e. isolated) HIV-1 protease. This is the next step to confirm AutoDock's prediction that they might "fit" and we learn from both correct and incorrect AutoDock predictions. Thank you again for your participation!
