New class of cancer drug halts tumor growth in early human trials

Preliminary results from a small human trial testing a novel cancer drug revealed over half the cohort had their tumors stop growing completely. The experimental drug, called berzosertib, is moving to larger clinical trials with experts expressing cautious optimism over this promising new treatment.

Berzosertib is part of an entirely new family of drugs designed to target a protein called ATR. This protein is known to play a fundamental role in repairing damaged DNA, and inhibiting its activity should hypothetically disrupt a tumor's ability to proliferate.

Phase 1 trials are not designed to investigate efficacy, instead they primarily focus on the safety and tolerability of a new drug in human subjects. However, as the study reports, the secondary anti-tumor endpoints seen in the trial were unexpectedly strong.

[Tested on severe ovarian cancer cases, may be applicable to other cancer types as well. More extensive human trials planned]

In a JAMA Viewpoint, Dr. Collins and Associate Director for Science Policy and Chief of Staff Dr. Carrie Wolinetz urge the scientific community to take concrete action to establish a Henrietta Lacks biospecimen consent policy that would require consent from research participants regardless of whether their biospecimens are identifiable. Honeybee venom found to be "extremely potent" against breast cancer

The research was conducted at Australia’s Harry Perkins Institute of Medical Research, where scientist Ciara Duffy has spent the last few years investigating the therapeutic potential of honeybee venom." We tested honeybee venom on normal breast cells, and cells from the clinical subtypes of breast cancer: hormone receptor positive, HER2-enriched, and triple-negative breast cancer,” Duffy explains.

The team found that a certain concentration of honeybee venom could be used to induce death in 100 percent of the cancer cells, while leaving healthy cells largely unharmed. Melittin, meanwhile, was found to “completely destroy cancer cell membranes within 60 minutes,” according to Duffy.

"We found both honeybee venom and melittin significantly, selectively and rapidly reduced the viability of triple-negative breast cancer and HER2-enriched breast cancer cells,” she says. “The venom was extremely potent.”

Synthetic DNA locks up drugs in the body – and only cancer has the key

The main ingredient is mucin, a protein found in the body in the form of mucus. This is held together with synthetic strands of DNA, and glycerol is added to keep the mucin particles from dissolving, and make them fold up around the active ingredient – the [anti-cancer] drug itself. Once it’s all together, the DNA strands won’t let the bundle unravel until they encounter the right “key” – a particular microRNA molecule.

In this case, the team chose microRNA that’s only found in cancer. These microRNA strands bind to the DNA and break down the bond holding the package together, releasing the drug. While many cells would take up the packages, the team says only diseased cells would be able to trigger the release of the drugs.

A common plant virus is an unlikely ally in the war on cancer

Promising results from injecting dog and mouse solid tumors with modified cowpea mosaic virus. When used in conjunction with existing treatments, increases cure rate and often eliminates recurrence of the cancer.

Common cold virus shows promise as cure for BCC, world's most often diagnosed skin cancer

A Queensland doctor believes he has found a way to cure patients with basal cell carcinoma (BCC) - the most common form of skin cancer - using the common cold virus, known as adenovirus. Dermatologist Greg Siller says he has achieved an 84 per cent “complete cure rate” of BCC in a trial where adenovirus is used as a vehicle to “deliver” cancer-fighting cells. [Series of four injections directly into the cancer] “We’re basically giving the cancer a big dose of the common cold,” Siller, an adjunct associate professor of the University of Queensland, told 7NEWS.

“We’re using adenovirus because it’s extremely effective in infecting cells. “The virus we use is live, but it’s been modified so it’s infertile. “We can deliver it directly into the cancer and it will replicate just once.
In that process, it delivers a human gene that codes for the protein that our immune system uses to destroy cancer cells.”

Siller believes the method can spark a “major change” in the treatment of BCC, as well as other cancers [can be used with existing drug treatments].

Drug-loaded nanoparticles dissolve only in cancer cells

“The highly selective toxicity of the particles made it possible for us to successfully treat two different types of highly aggressive pleural tumors in mice,” says Hanna Engelke, co-author of the study. “With only two doses, administered locally, we were able to reduce tumor sizes by 40 and 70 percent, respectively. Clearly, the particles can be highly toxic to cancer cells. Indeed, we found that the more aggressive the tumor, the greater the killing effect.”
