"Structure discovered of cancer drug candidate

Chemists at The Scripps Research Institute (TSRI) have determined the correct structure of a highly promising anticancer compound approved by the U.S. Food and Drug Administration (FDA) for clinical trials in cancer patients.

The new report, published this week by the international chemistry journal Angewandte Chemie, focuses on a compound called TIC10.

In the new study, the TSRI scientists show that TIC10's structure differs subtly from a version published by another group last year, and that the previous structure associated with TIC10 in fact describes a molecule that lacks TIC10's anticancer activity.

By contrast, the correct structure describes a molecule with potent anticancer effects in animals, representing a new family of biologically active structures that can now be explored for their possible therapeutic uses.

"New anticancer compound discovered using accelerated drug screening process

A team of research scientists from VTT Technical Research Centre of Finland, the University of Turku and the University of Eastern Finland has discovered a previously unknown Cent-1 molecule that kills cancer cells. Their research also shows that new cancer drug candidates can be identified faster and at lower cost by using computer-assisted and cell-based screening of compounds.

The objective of the research project led by Marko Kallio, Principal Scientist at VTT, was to accelerate the drug development process by identifying new compounds that would possess similar binding properties and cellular phenotype , but a different chemical structure, as the selected drugs in clinical use or investigational compounds in development.

The scientists combined computer-based screening and cell-based assays to create a method that can significantly accelerate drug discovery and thereby lower development costs. It is highly likely that the new compounds identified using this method have not yet been patented.

The research team conducted a computer-assisted screening of 65,000 compounds and cell-based assays on the 150 highest scoring hit compounds, before identifying the Cent-1 molecule. The Cent-1 molecule kills cancer cells through a mechanism similar to that of the template drug Rigosertib that is currently under commercial development. However, since the chemical structure of the Cent-1 compound differs from Rigosertib, there are no major obstacles to further development....."

"Drug discovery accelerates immunotherapy for cancer

Researchers at UCLA's Jonsson Comprehensive Cancer Center have discovered a drug that enhances an immune-based treatment for cancer. Dr. Robert Prins, associate professor of neurosurgery and molecular and medical pharmacology and Dominique Lisiero, a PhD student in the Prins laboratory, led the research.

The study was published in the April edition of the Journal for Immunotherapy of Cancer.

The treatment uses transplantation of T cells, the foot soldiers of the immune system, in a process called T cell adoptive transfer, that are trained specifically to attack cancer cells transplanted into animals. The new drug, a histone deacetylase inhibitor called LBH589 (generic name: panobinostat), was found to enhance the activity of the T cells and of the immune system's inflammatory response in mice, giving a boost to the treatment's tumor-shrinking effect.

With the drug, the transferred T cells multiplied faster, survived longer and rejected the established cancer more efficiently...."

"Green tea component upsets cancer cell metabolism

A new study reveals how an active component of green tea disrupts the metabolism of cancer cells in pancreatic cancer, offering an explanation for its effect on reducing risk of cancer and slowing its progression. The researchers believe the discovery signals a new approach to studying cancer prevention.

Reported in the journal Metabolomics, the study explores the effect of epigallocatechin gallate or "EGCG," an active biological agent of green tea. It shows that EGCG changes the metabolism of pancreatic cancer cells by suppressing the expression of lactate dehydrogenase A or LDHA, a critical enzyme in cancer metabolism.

Metabolism is all the chemical reactions that occur in cells - such as extracting and using energy - that keep them alive, growing and multiplying. These cells can be normal, and they can also be cancerous.

Numerous studies have previously suggested green tea and its extracts may provide suitable treatments for cancer, as well as other diseases.

For example, one published in 2012 suggested that drinking green tea may lower risk of digestive system cancers in women while another found EGCG delivered intravenously directly to tumors made two-thirds of them shrink or disappear within one month..."

Statins are typically used to lower blood cholesterol levels by blocking a chemical in the liver necessary for making cholesterol. But new research presented at the 50th Annual Meeting of the American Society for Clinical Oncology suggests patients in late stages of cancer or other terminal illnesses may extend their lives by discontinuing use of statins....."

"The transcription factor STAT3 mediates tumor survival, proliferation, invasion, and immunosuppression, and is persistently activated in tumor cells and tumor-associated immune cells.

In T cells, STAT3 activation impairs anti-tumor responses, while inhibition or loss of STAT3 promotes anti-tumor responses.

In this issue of the Journal of Clinical Investigation, Hua Yu and colleagues at the Beckman Research Institute developed an aptamer-based strategy to deliver STAT3 siRNA and inhibit STAT3 in tumor cells and tumor-associated immune cells.

An aptamer that binds the cell surface receptor cytotoxic T lymphocyte-associated antigen-4 (CTLA), an inhibitor of the T cell anti-tumor response, was attached to STAT3 siRNA (CTLA4apt-STAT3 siRNA), allowing for targeted delivery to tumor-associated T cells, Tregs or malignant T cells....."

"Our own treacherous immune genes can cause cancer after viral infection

HPV (human papillomavirus) infection is widely known to induce cancer. Many of the mutations that cause this virally-induced cancer are caused by a family of genes that normally combats viral infections, finds new UCL research.

This raises the possibility of developing drugs to regulate the activity of these genes to prevent HPV-associated cancers from developing and reduce the ability of existing cancers to evolve resistance to treatments.

The research, published in Cell Reports, shows for the first time that genes from the 'APOBEC' family, which help to fight off viral infection, actually cause mutations that lead to HPV-associated cancer. This research was funded by the Rosetrees Trust, a charity dedicated to supporting pioneering medical research, with additional funding from the Debbie Fund and Cancer Research UK.

"Genes from the APOBEC family encode proteins that modify the DNA of invading viruses, causing mutations that prevent the virus from replicating," explains senior author Dr Tim Fenton, of the Tumour Virus team at the UCL Cancer Institute. "We now provide evidence that they can also cause mutations in our own DNA after HPV infection, leading to cancer."

"Prolonged fasting 're-boots' immune system

Results of a new study on mice and a phase 1 trial of humans suggest that prolonged cycles of fasting - for 2-4 days at a time - not only protect against toxic effects of chemotherapy, but also trigger stem cell regeneration of new immune cells and clearing out of old, damaged cells.

The study, by researchers from the University of Southern California (USC) in Los Angeles, and published in the journal Cell Stem Cell, is the first to show that a natural intervention can trigger regeneration of an organ or system through stem cells.

The team believes the findings could benefit people with immune system damage, for example if they have received chemotherapy treatment for cancer. It could also benefit the elderly whose immune systems are weakened through aging, making them more susceptible to disease.

The scientists say prolonged fasting appears to shift stem cells of the immune system from a dormant state to an active state of self-renewal.

Results from experiments with mice and a phase 1 human clinical trial showed that long periods of fasting significantly lowered levels of white blood cells. In the mice, it flipped a switch that changed the signaling pathways of hematopoietic stem cells - a group of stem cells that generate blood and immune systems.

"We could not predict that prolonged fasting would have such a remarkable effect in promoting stem cell-based regeneration of the hematopoietic system," says Valter Longo, a professor of Gerontology and the Biological Sciences at the USC Davis School of Gerontology, and director of the USC Longevity Institute.

He says that when you stop eating, the body uses up stored glucose, fat and ketones, and also recycles worn out and damaged immune cells..."

"Australian scientists treating the most common form of childhood cancer have made an important breakthrough in understanding what drives it to grow.

Professor Glenn Marshall from the Children's Cancer Institute and his team have discovered a gene linked to the cause of neuroblastoma that could provide new targets for cancer therapy. The team's work has shown that junk DNA is involved in causing neuroblastoma.

Dr Tao Lui from the Children's Cancer Institute discovered that a new long non-coding RNA plays an important part in the formation of neuroblastoma tumours."
